{"id":182,"date":"2010-05-02T19:21:24","date_gmt":"2010-05-02T17:21:24","guid":{"rendered":"http:\/\/www.pjh.fr\/wordpress\/?page_id=182"},"modified":"2015-06-15T17:23:59","modified_gmt":"2015-06-15T15:23:59","slug":"regulation-de-production-depo","status":"publish","type":"page","link":"http:\/\/www.pjh.fr\/wordpress\/?page_id=182","title":{"rendered":"R\u00e9gulation de production d&rsquo;EPO"},"content":{"rendered":"<p>La r\u00e9ponse cellulaire\u00a0 \u00e0 l&rsquo;hypoxie est contr\u00f4l\u00e9e par une famille de facteurs de transcription : <strong>hypoxia-inducible factors (HIFs)<\/strong>.<\/p>\n<blockquote><p>HIF est un h\u00e9t\u00e9rodim\u00e8re compos\u00e9 de deux sous-unit\u00e9s : HIFalpha\u00a0et HIFalpha.<br \/>\nHIFalpha existe sous la forme de 3 isoformes (HIF1alpha, HIF2alpha et HIF3alpha) qui sont synth\u00e9tis\u00e9es de mani\u00e8re continue en r\u00e9ponse \u00e0 l&rsquo;hypoxie.<\/p><\/blockquote>\n<p><strong>HIFs r\u00e9gulent la transcription du g\u00e8ne de l&rsquo;EPO<\/strong> et donc la synth\u00e8se d&rsquo;EPO qui est produite essentiellement par des cellules p\u00e9ritubulaires r\u00e9nales. HIF2alpha est le principal isoforme r\u00e9gulant l&rsquo;expression de l&rsquo;EPO.<\/p>\n<p><em><strong>En situation de normoxie<\/strong><\/em>, HIFalpha est d\u00e9truit rapidement par l&rsquo;effet combin\u00e9 de l&rsquo;oxyg\u00e8ne, d&rsquo;enzymes \u00e0 domaine prolyl hydroxylase (PHD) et la prot\u00e9ine VHL (Von Hippel-Lindau), suppresseur de tumeur. La forme hydroxyl\u00e9e de HIFalpha peut se fixer \u00e0 VHL et le complexe HIFalpha-VHL facilite la d\u00e9gradation de HIFalpha via le prot\u00e9asome (Cf. Figure).<\/p>\n<p><em><strong>En situation d&rsquo;hypoxie,<\/strong><\/em> la d\u00e9gradation prot\u00e9asomale de HIFalpha est ralentie entra\u00eenant son accumulation au niveau cytoplasmique, son passage au niveau nucl\u00e9aire et sa dim\u00e9risation avec HIFalpha. Les deux mol\u00e9cules ensemble se lient \u00e0 de courtes s\u00e9quences d&rsquo;ADN qui contiennent les \u00e9l\u00e9ments de r\u00e9ponse \u00e0 l&rsquo;hypoxie et qui r\u00e9gulent la transcription oxyg\u00e9no-d\u00e9pendante des g\u00e8nes cibles de HIF.
